FA Carlsberg Vase holders Truro City will begin the defence of their title away at Almondsbury Town after the draw for the Second Round proper was made at Soho Square on Monday.
The Cornish outfit, who beat AFC Totton at Wembley in May, will play their first Vase game of the season against the Hellenic League side at the Oaklands Park Stadium.
Meanwhile, AFC Totton will welcome VCD Athletic to Testwood Park for thier tie, which is schedule for 17 November.
The First Round's biggest winners, Desborough Town, who thrashed Bugbrooke St Michael 9-0 at home on Saturday, face a tricky away trip to Suffolk where they will take on Lowestoft Town.
There will be 64 ties in all with £1,000 up for grabs from The FA's prize fund for each of the winning sides.
