The oldest club in the world, Sheffield FC, produced a stirring second-half fightback as the team founded in 1857 came from three goals down at half-time to earn a replay against Guiseley.
Sutton United revived some giant-killing memories from two decades ago as they came from a goal down with ten minutes left to overcome Ware 2-1.
One of last year's giant-killers Chasetown lost 2-0 to Evesham after conceding two late goals.
But another team with fond memories of last year's competition will be in the hat for the next round as Havant & Waterlooville beat Godalming Town 2-1.
Nantwich Town scored four second-half goals to record an impressive 5-1 win at Whitley Bay.
Durham City went one better, beating Stalybridge 6-1 away despite going behind to a fourth minute strike. Gavin Cogden notched a hat-trick for the north-easteners.
Ben Walker was another hat-trick hero as he helped Belper Town to a 4-1 win over Prescot Cables.
Maidstone United continued their good run in this season's competition with a 4-1 win at Halesowen.
One of the most eagerly-anticipated ties saw Dover and AFC Wimbledon, both flying high in their respective divisions, play out a goalless draw.
Bishop's Stortford squeezed through to the next round 2-1 against Rugby Town courtesy of a last-minute penalty.
Frome Town nearly upset Team Bath, but the Conference South side scored two second-half goals to come away with a 2-2 draw.
Elsewhere there were wins for Hampton & Richmomd, AFC Telford, Hosrham, Hinckley United and Blyth Spartans.
