Winning clubs will receive £30,000 from The FA
Three non-League sides upset the odds and progressed to the Third Round on Saturday where a possible meeting with a Premier League giant could be on the cards.
However, Droylsden were cruelly denied the chance of a place in the Third Round themselves after their clash with Chesterfield was postponed at half-time - with the Conference North side leading their League Two opponents.
Carl Lamb met Neil Prince's cross to head the visitors into a shock lead ten minutes from the interval but heavy fog at Saltergate means they will have to try their cup heroics one more time.
Elsewhere, Fleetwood almost followed Eastwood's shining example but the Conference South side were heartbroken by League One side Hartlepool.
Andy Bell gave the non-league outfit a stunning lead but Michael Mackay met Jamie McCunnie's cross to level it. Mackay added his second of the game after eleven seconds of the second half had elapsed and a minute later Joel Porter made it three as the floodgates threatened to open.
But Adam Warlow hit a late goal to make it a nervous finale for the visitors, but Pools held on to kill off Fleetwood's dream.
Fellow non-League side Alfreton Town didn't fair much better after Scunthorpe cruised to a 4-0 win courtesy of goals from Ben May, Gary Cooper (2) and Sam Togwell.
Danny Graham and Graham Kavanagh were on target as Carlisle beat Grays 2-0 but the romance of The FA Cup was rekindled again down at Forest Green.
The Conference Premier stugglers beat high-flying League Two side Rochdale, who were unbeaten in the last nine league games. But Jonathan Smith and Josh Low sank Dale to book their place in the hat for the Third Round draw and a date with the big boys.
Jason Demetriou and Danny Granville netted as Leyton Orient beat Bradford 2-1 at Valley Parade. Michael Boulding had equalised for the hosts but Martin Ling's men got back in front and held on to advance to the next round.
Kenny Jackett's improving Millwall side made short work of Aldershot - Gary Alexander's double and a strike from Ashley Grimes was enough for the League One side to triumph at the New Den.
Gillingham and Stockport will have to do it all again after it ended goalless at Priestfield Stadium, as did Peterborough's game with Tranmere.
But at least they're in draw for the Third Round - and they will be joined by Southend after they beat Mick Harford's Luton 3-1 and Torquay, after their 2-0 win at home to Oxford.
In the one remaining First Round proper tie in Essex, Grays Athletic entertained Carlisle United, but it was the Cumbrians who were doing the scoring.
A goal in each half from Danny Graham and Graham Kavanagh finally ended the tie and lined up the Second Round meeting at home with Crewe Alexandra on Wednesday night.
